Information on Enzyme Antibody Conjugates
Enzyme conjugated affinity purified antibodies are useful
in immunohistochemistry, Elisa procedures, Western and other immunoblotting
assays and immunoelectronmicroscopy. PROTOS IMMUNORESEARCH offers
primary and secondary antibodies conjugated to peroxidase or alkaline
phosphatase. The working dilutions for these reagents are 1:1,000
to1:5,000 for peroxidase, and 1:1,000 to 1:3,000 for alkaline phosphatase,
though these values vary with individual modifications of the techniques.
Studies with enzyme conjugates of affinity
purified antibodies sometimes are complicated by the non-specific
binding of the Fc part of immunoglobulins by Fc-receptors on tissues
and cells.This complication can be eliminated by enzymatically removing
the Fc portion of the antibody before preparing the enzyme conjugate.
PROTOS IMMUNORESEARCH provides enzyme conjugates of F(ab')2 fragments
with peroxidase or alkaline phosphatase. The working dilutions of
these reagents are approximately the same as for the enzyme conjugates
of the intact molecules.
For applications where tissue or
cellular penetration of antibody-enzyme conjugates is important, PROTOS
IMMUNORESEARCH offers low molecular weight Fab'
antibody(i.e.3.5S) fragments conjugated to peroxidase.
